Introduction

PARACETAMOL

XX is analgesic and antipyretic agent. XX is the active metabolite of phenacetin, responsible for its analgesic effect. XX is a weak prostaglandin inhibitor in peripheral tissues and possesses no significant antinflammatory effects. XX is one of the most important drug used for the treatment of mild to moderate pain when an antinfalmmatory effect is not necessary. XX is preferred over aspirin as an analgesic/antipyretic for patients in whom aspirin is contraindicated, such as those who have a history of gastric ulcer or a coagulation disorder.

CODEINE

XX is the 3-methyl ether of morphine. It can be obtained from opium, present in a concentartion of less than 0.5% or synthesized by methylation of morphine.a narcotic analgesic (relieves the pain without causing sleep). XX is used to relieve mild to moderate pain. it is also used as a cough suppressant.

Paracetamol also known as Acetylparaminophenol, Acetaminophen is of Synthetic origin and belongs to Aminophenol. It belongs to Cyclo-oxygenase inhibitor pharmacological group on the basis of mechanism of action and also classified in Analgesics and Anti-inflammatory Agents pharmacological group.The Molecular Weight of Paracetamol is 151.20.
Its pKa is 9.5.

Codeine also known as Codeine (Phosphate) is of Semi Synthetic origin and belongs to Opioid. It belongs to OPIATE agonist pharmacological group on the basis of mechanism of action and also classified in Analgesic pharmacological group.The Molecular Weight of Codeine is 406.40.
It is weakly acidic drug, 7.29% solution of the drug is isotonic and Its pKa is 8.2.

Pharmacokinetics

Oral absorption of Paracetamol is found to be 96.5% ±1.5. Volume of distribution is found to be 0.9 l/kg and plasma protien binding is < 20%. Presystemic metabolism is noted to be 20% and metabolism is reported hepatic. Renal Excretion accounts for 5% unchanged and plasma half life is 1.5-3.0 hr.

Oral absorption of Codeine is found to be 94% ±4. Volume of distribution is found to be 3.6 l/kg and plasma protien binding is 7-25%. Presystemic metabolism is noted to be 50% and metabolism is reported hepatic. Renal Excretion accounts for major and plasma half life is 3-4 hr.
